Moving beyond tamoxifen: Drug discovery and the future of selective hormone receptor modulators

The transformation of tamoxifen, from cast-off to lifesaver, laid the foundation for a new class of therapeutics -- selective estrogen receptor modulators. V. Craig Jordan, a researcher at the Fox Chase Cancer Center in Philadelphia, Jordan reports on efforts to use the lessons from tamoxifen to develop new hormone receptor-related drugs for both women and men.

Researchers at the University of California San Diego have developed a novel computer technique to search for the side effects of major pharmaceuticals. The study, reported Nov. 30 in PLoS Computational Biology, relates to a class of drugs known as Select Estrogen Receptor Modulators, which includes tamoxifen, the most prescribed drug in the treatment of breast cancer.

New data from the National Surgical Adjuvant Breast and Bowel Project (NSABP) B-33 study, presented today at the San Antonio Breast Cancer Symposium, showed that postmenopausal women with hormone receptor-positive breast cancer who received Aromasin after five years of tamoxifen were 56 percent less likely to have a relapse of breast cancer than those who received placebo.
