Researchers at Mayo Clinic Cancer Center, in cooperation with industry partners, have, for the first time, identified tumor specific alterations in the cellular pathway by which the multiple myeloma drug bortezomib (Velcade) works, and they have identified nine new genetic mutations in cancer cells that should increase a patient's chance of responding to the agent.

A group at Hiroshima University Hospital performed a matched historical controlled study on whether a 24-week course of interferon-alpha therapy, after curative treatment for primary hepatocellular carcinoma associated with hepatitis C virus, could influence tumor recurrence, patient survival, and liver function. Patients with sustained virological responses had reduced recurrence, prolonged survival and a preserved liver function. The group also determined that viral eradication was the most important factor in such patients.

Tumor-reactive T cells boosted by hematopoietic stem cell transplantation

Treatment for skin cancer by infusion of tumor-reactive T cells requires patients to be pre-treated with agents that transiently decrease the number of immune cells (nonmyeloablative agents). A study in mice now indicates that pre-treatment with more intense immune cell-depleting strategies (known as myeloablative strategies) and a hematopoietic stem cell transplant enables infused tumor-reactive CD8+ T cells to increase in number more than pre-treatment with nonmyeloablative agents, and this correlates with increased tumor regression.

Counting circulating tumor cells before and after the start of treatment for patients with metastatic colorectal cancer could help doctors determine when or if a change in treatment should be made. The results of a large, multicenter study laying the groundwork for such decisions will be presented at the annual meeting of the American Society of Clinical Oncology. The study showed the number of circulating tumor cells is a significant predictor of survival and cancer progression.

A multi-center team, led by Vanderbilt-Ingram Cancer Center investigators, has developed a new pre-treatment blood test that predicts which non-small-cell lung cancer patients will live longer when they are treated with certain targeted cancer therapies (Iressa, Tarceva). The mass spectrometry-based test, described in the June 6 Journal of the National Cancer Institute, may allow physicians to select the most beneficial therapy for each lung cancer patient, a step forward in the era of personalized medicine.
