Role of TRPM8 in the development of prostate cancer
05-17-2007 · EurekAlert!In normal prostate epithelium, cells coexist in many stages of development-differentiation, and disrupted differentiation and proliferation are major causes of cancer. A new study shows that the endoplasmic reticulum protein TRPM8 (ER-TRPM8) present in human prostate epithelial luminal cancer cells retains it’s function as a calcium release channel, independent of the differentiation state of the cell, and may be an important factor in controlling the growth of prostate cancer cells.

New Phase II data presented today suggest that ZD4054, a novel compound in development for the treatment of men with hormone-resistant prostate cancer, could offer a promising improvement in overall survival in men with metastatic HRPC who were asymptomatic or mildly symptomatic for pain.1 Patients who received ZD4054 10mg once-daily experienced a 45 percent reduction in the risk of death compared to placebo.

The results from a randomized, double-blind, placebo-controlled, Phase II study of ZD4054 -- a specific endothelin A receptor antagonist -- were delivered today at the 14th European Congress of Clinical Oncology (ECCO, 23-27 Sept., Barcelona). The data presented today for ZD4054, an investigational compound in development for the treatment of men with hormone-resistant prostate cancer, showed no differences in progression-free survival but did report encouraging overall survival data.
